Lund DK, Mouly V, Cornelison DD. MMP-14 is necessary but not sufficient for invasion of three-dimensional collagen by human muscle satellite cells. Am J Physiol Cell Physiol 307: C140–C149, 2014. First published June 4, 2014; doi:10.1152/ajpcell.00032.2014.—The twenty-five known matrix metalloproteases (MMPs) and their endogenous inhibitors, tissue inhibitors of metalloproteases (TIMPs), mediate cell invasion through the extracellular matrix (ECM). In a comparative three-dimensional assay, we analyzed human and mouse satellite cells’ competence to invade an artificial ECM (collagen I). We identified a single MMP that 1) is expressed by human muscle satellite cells; 2) is induced at the mRNA/protein level by adhesion to collagen I; and 3) is necessary for invasion into a collagen I matrix. Interestingly, murine satellite cells neither express this MMP, nor invade the collagen matrix. However, exogenous human MMP-14 is not sufficient to induce invasion of a collagen matrix by murine cells, emphasizing species differences.

متن کاملStudy of Human Chondrocyte Redifferntiation Capacity in Three-Dimensional Hydrogel Culture
Objective(s) Articular cartilage tissue defects cannot be repaired by the proliferation of resident chondrocytes. Autologous chondrocyte transplantation (ACT) is a relatively new therapeutic approach to cover full thickness articular cartilage defects by in vitro grown chondrocytes from the joint of a patient. Therefore, we investigated the redifferentiation capability of human chondrocytes ma...
متن کاملCostal Versus Articular Chondrocytes in Alginate Three-Dimensional Cultures
Given the difficulties in accessing articular cartilage as a source of chondrocytes to be used in fabricating cartilage constructs, alternative sources are required. The present study examined chondrocytes from costal cartilage for their suitability in cartilage tissue engineering. Chondrocytes isolated from rat knee and rib hyaline cartilage were separately mixed with alginate and placed in a ...
متن کاملIsolation and optimization of mice skeletal muscle satellite cells using preplating method and culture media substitution
Introduction: Satellite cells are known as the main regenerative cell type in skeletal muscles. Our study established a modified digestion and preplating method for the isolation of slow or weak adherent cells for the enrichment of satellite cells. Low-survival rate of these primary stem cells prompted us to address whether cell culture medium substitution might change cell viability status. M...
متن کاملWharton’s Jelly Mesenchymal Stem Cell: Various Protocols for Isolation and Differentiation of Hepatocyte-Like Cells; Narrative Review
There are several differentiation methods for mesenchymal stem cells (MSCs) into hepatocyte-like cell. Investigators reported various hepatic differentiation protocols such as modifying culturing conditions or using various growth factors/cytokines. In this literature review, we compared different MSCs extraction and isolation protocols from Wharton’s jelly (WJ) and explored various MSCs differ...
